The BBB is formed by a complex cellular system of endothelial cells, astroglia, pericytes, perivascular macrophages and a basal membrane, although the anatomic substrate of the BBB is the interendothelial tight junctions that form a continuous sealing. The BBB serves as an exquisitely controlled, functional gate to the eNS. It not only protects the brain from agents in the blood that could impair neurological function, but also controls the influx and efflux of numerous substances to maintain proper homeostasis and provide the brain with necessary nutrients. The structural and functional integrity of the BBB was shown to be dramatically altered during various diseases of the eNS, including neoplasia, ischemia, trauma, hypertension, inflammation and epilepsy. Recent years research has partially elucidated the mechanisms underlying the development of some of these brain disorders as well as the pathways used by different pathogens, like bacteria and viruses, to initiate eNS infections. The development of in vitro models of the BBB had instrumental role in the understanding of the involvement of the BBB in the pathogenesis of several eNS diseases. The venue of the 20th meeting was the prestigious, fully restored Schloss Waldthausen (Waldthausen Castle) which provided a special setting for ISOTT 1992. The beautiful front view of the castle became part of the ISOTT 1992 logo. The 20th ISOTT Meeting was held in Mainz from August 26th through August 30th, 1992. The Conference attracted 200 active participants from 16 countries. The theme of this meeting emphasized oxygen transport to tumors but as in earlier meetings, essentially all aspects of oxygen transport within the body were covered as demonstrated by the manuscripts comprising this volume of the series 'Oxygen Transport to Tissue'. All manuscripts were reviewed. Extensive revisions were made in about 25% and modest revision in about another 30%. ' Among various kinds of energy transduction mechanisms in biological systems, the mechanism of muscle contraction has been studied most intensively and extensively over many years. Since the monumental discovery by the two Huxleys and coworkers that muscle contraction results from relative sliding between the thick and thin myofilaments, attention of muscle investigators has been focused on the question, what makes the fllaments slide past one another. In response to the above question, A. F. Huxley and Simmons put forward a contraction model in 1971, in which globular heads of myosin (cross-bridges) extending from the thick fllament first attach to actin on the thin fllament, and then change their angle of attachment to actin (power stroke) leading to force generation or myofilament sliding until they detach from the thin fllament. The rocking cross-bridge contraction model seemed to be entirely consistent with the kinetic scheme of actomyosin ATPase published by Lymn and Taylor at the same time, thus giving a strong impression to the people concerned that the muscle contraction mechanism would soon be sorted out.
